چکیده

Whether and how DNA methylation influences the binding of transcription factors (TFs) to their corresponding DNA sequence motifs in vivo remains largely unresolved. In a recent publication, Schübeler and coworkers (Domcke et al, 2015) identify a few methylation-restricted TFs in mouse embryonic stem cells, including NRF1. The authors show that NRF1 binding to its motif can be outcompeted by de novo DNA methylation, suggesting that methylationsensitive TFs rely on neighbouring motifs in cis—bound by pioneer TFs—to ensure local hypomethylation.

Evaluation of MYB93 and MAD8 Genes in Transgenic and Non-Transgenic Rice

Increasing drought tolerance, especially in rice, which is one of the most important crops in Asia, is necessary. Transcription factors are specific sequence DNA-binding proteins that are capable of activating or suppressing transcription. These proteins regulate gene expression levels by binding to cis regulatory elements in the promoter of target genes to control various biological processes ...

DNA hypomethylation and germ cell-specific expression of testis-specific H2B histone gene.

Testis-specific H2B (TH2B) histone gene of rat is expressed during meiotic event of spermatogenic differentiation. The gene is unusual in that it has conserved the regulatory elements involved in the S phase-specific transcription of somatic H2B genes as well as the S phase-specific stabilization of histone mRNA. Genomic sequencing revealed that all analyzed CpG sites in the promoter region of ...

p53 binding sites in normal and cancer cells are characterized by distinct chromatin context

The tumor suppressor protein p53 interacts with DNA in a sequence-dependent manner. Thousands of p53 binding sites have been mapped genome-wide in normal and cancer cells. However, the way p53 selectively binds its cognate sites in different types of cells is not fully understood. Here, we performed a comprehensive analysis of 25 published p53 cistromes and identified 3,551 and 6,039 'high-conf...

DNA methylation profile of tissue-dependent and differentially methylated regions (T-DMRs) in mouse promoter regions demonstrating tissue-specific gene expression.

DNA methylation constitutes an important epigenetic regulation mechanism in many eukaryotes, although the extent of DNA methylation in the regulation of gene expression in the mammalian genome is poorly understood. We developed D-REAM, a genome-wide DNA methylation analysis method for tissue-dependent and differentially methylated region (T-DMR) profiling with restriction tag-mediated amplifica...

گروه بندی و بررسی الگوی بیان ژن های خانواده bZIP در ریشه گیاه گوجه فرنگی تحت تنش دمای پایین

Transcription factors (TFs) are master regulators that control gene clusters Plant bZIP (basic region/leucine zipper) transcription factors play crucial roles in biological processes. The Tomato genome sequence contains 73 genes of bZIP transcription factors.  The bZIPs in tomato have never been classified. In this study, 73 genes of bZIP transcription factors were classified in 11 groups by th...
